BD Analyzers
General Rules
- Remember to always check the printed schedule at the beginning and end of your appointment; you need to know if you are either the first or last user of the day. If you are the first user of the day you must prime and perform the bead QC procedure (QC procedure only required for FACScan and FACSCalibur instruments). If you are the last user of the day you are required to turn off the instrument and shut down the computer.
- After performing bead QC, place beads back in 4°C fridge. DO NOT leave the beads out for extended periods of time. NEVER leave beads on the SIP arm in standby mode for long periods of time as this will throw-off the appropriate bead concentration.
- When making up fresh APC beads (for FACSCalibur bead QC) please follow these instructions:
- Obtain the APC beads manufacturer's bottle and shake well
- Add ONE drop of beads from the APC beads bottleto 0.5ml of sheath fluid/isoflow.
- After you run your samples and perform the required cleaning, be sure to empty the waste tank and fill the sheath fluid tank. The waste tank is to be emptied in the biohazard sink (at the back of lab, farthest from facility entrance, NOT down the one closest to door).
- If there is greater than a two-hour time slot between your appointment and the next appointment please turn off both the instrument and computer.
Data Storage
- Storage space on the equipment has limitations and needs to be accessible to everyone. Maintenance and data storage operations are performed on a regular basis:
- FACScan and FACSCalibur instruments: all files acquired within a month (excluding CellQuest Pro templates and instrument settings) are erased on the second Monday of the following month (example: on the second Monday of September, all data acquired in August is deleted from the hard drive).
- LSRII instruments: data will be deleted from DIVA software by staff every two weeks. Data and experiment templates can be temporarily exported to the D://BDExportFCFusers only. Data or experiments located in the D:// drive will be deleted every 1st and 3rd Monday of the month. Data placed in any other location will be deleted.
