Cell Sorters

The facility houses three FACSArias (Becton Dickinson), one MoFlo (Dako Cytomation), and one Influx (BD-Bioscience). Users will need to book time with FCF staff as these instruments are operated by FCF staff exclusively.

Our three FACSArias have different laser and PMT configurations. The number of fluorescence PMTs per laser and the laser excitation wavelength and power is shown below for each sorter:

Aria-RITT (11 colours: 6B/2R/3V):
488 nm Blue (20 mW)
633 nm Red (18 mW)
407 nm Violet (15 mW)

Aria-GC (13 colours: 7B/3R/3UV):
488 nm Blue (100 mW)
633 nm Red (30 mW)
355 nm UV (20 mW)

Aria-SC (13 colours: 7B/3R/3V):
488 nm Blue (15 mW)
633 nm Red (15 mW)
407 nm Violet (10 mW)

The filter/PMT configurations are flexible. Please consult the Fluorochrome Excitation/Emission Table and Laser/Filter Combination Table to design your experiment. Check with FCF Staff to very that we have the filters you need for your experiment.

The sheath pressure for sorting can be varied between 20-60 PSI, with flow rates varying from 10,000-20,000 events per second, depending on nozzle size (70 µm or 100 µm) and the specific parameters of your sample.

Note: Our typical laser configuration for the Mo-Flo is 100 mW of 488 nm from the iCyt, 100 mW of 647 nm 100 mW from the Spectrum, and 50 mW of multiline UV from the 302c laser. The power (mW) outputs from the tunable laser will vary according to the excitation wavelength. Please consult with FCF staff if you need further information.
