Attention LSRII users:

In an effort to standardize and simplify the use of the BD analyzers, by March 22 all LSRII analyzers will have one single cytometer configuration called "New Advanced Configuration". This configuration was designed to accommodate the most number of commonly used fluorochromes and is essentially identical to the current default configuration on the LSRIISC, LSRIICFI and LSRIIGC instruments. (LSRIIOICR users will be contacted regarding changes to this instrument).

Starting from March 8 the new default configuration will be this "New Advanced Configuration" which means that if you changed the cytometer configuration and any of the filters, the instrument must be returned to this configuration at the end of your appointment. (As always, additional filters can be signed out from the FCF office the day before your experiment.)

Note that the old configurations will still remain. However, on March 22 the old configurations will be deleted. This means that CS&T voltages will only be available for this configuration. Users are reminded that while CS&T voltages provide reasonable starting PMT voltages for many applications they may not be truly optimal for your application since these voltages were established using beads and not cells.

Users who wish to use filters that are not part of this configuration can easily establish their own optimal voltages. The protocol to do this is posted on our website under "Basic Protocols". We will be going over this protocol at the FCF User's group meeting on Monday Feb 22, 1-3 p.m., room 14-203.
