Method : NEFA is a colorimetric method.
The Randox enzymatic method relies upon acylation of coenzyme A (CoA) by the fatty acids in the presence of acyl-CoA synthetase (ACS). The acyl-CoA produced in the reaction is further oxidized by acyl-CoA oxidase (ACOD) with generation of hydrogen peroxide. Hydrogen peroxide, in the presence of peroxidase (POD) permits the oxidative condensation of N-ethyl-N-(2hydroxy-3-sulphopropyl)-m-toluidine (TOOS) with 4-aminoantipyrine (4-AAP) to form a purple coloured adduct which can be measured at 548 nm.
